Semaglutide is a synthetic GLP-1 receptor agonist with structural modifications that dramatically extend its half-life to roughly one week. That long half-life is what enables once-weekly injectable dosing and a stable receptor signal between doses.
Mechanism of action
Semaglutide binds the GLP-1 receptor and mimics the effects of endogenous GLP-1: glucose-dependent insulin secretion, glucagon suppression, delayed gastric emptying, and centrally mediated appetite reduction. Unlike native GLP-1, it resists degradation by DPP-4 and stays active for days.
